Study on the Determination of Dilinoleoylphosphatidylcholine Concentration in Human Plasma with High Performance Liquid Chromatography-Mass/Mass

ABSTRACT
Objective To study the concentration of dilinoleoylphosphatidylcholine(DLPC) in healthy volunteers with high performance liquid chromatography-mass/mass.Methods From November 2010 to July 2011,7 volunteers were included in our research.Three of them took the Polyene Phosphatidyl choline soft capsule(test formulation),three took Polyene Phosphatidyl choline capsule(reference formulation),and the other one did not take any medicine.We collected the plasma of these volunteers and explored different determined methods and sample pretreatment methods.The established method was used to determine the concentration of DLPC in human plasma.Results The study was conducted with an API 3000 HPLC-MS/MS system consisted of an Ultimate CN column(50 mm×4.6 mm,5 μm).The mobile phase consisted of methanol-water-formic acid(80︰20︰0.05,V/V/V) at a flow rate of 0.5 mL/min.Gliclazide was used as the internal standard.The sample was operated under the multiple reaction monitoring mode using the APCI in positive mode.After taking oral test formulation and reference formulation of DLPC capsule,plasma concentrations of DLPC had no significant changes.Plasma concentration of DLPC of the volunteer who did not take the capsule was also at a high level.Conclusion This method for quantitative determination of DLPC can not be applied in its bioequivalence.
